Retatrutide is a weekly injection that is slightly more sophisticated than other GLP-1 drugs because it is a “triple agonist.” Unlike current drugs (Zepbound, Ozempic-type medications) that target one or two hormone pathways, retatrutide activates three hormone receptors (GIP, GLP-1, and glucagon) at once
TRIUMPH had separate clinical trials: one in individuals with type-2 diabetes and obesity, and the other in non-diabetics with obesity and cardiovascular disease

TRIUMPH-2 featured about 1,150 participants with type-2 diabetes and obesity. At the highest dose (12 mg), participants lost an average of 50 lbs (20.8% of body weight) over 80 weeks. Blood sugar control (A1C) also improved significantly. A1C is a measure of blood sugar over a period of time, rather than an instantaneous reading, and shows more steady results
TRIUMPH-3 featured about 1,950 participants with obesity and cardiovascular disease. The highest dose led to an average loss of 56 lbs (22.6% of body weight) over 80 weeks. This is among the largest weight-loss results reported in a trial of this size. As for heart disease risk markers: triglycerides dropped 37%, waist circumference shrank about 7.5 inches, blood pressure and inflammation markers (hsCRP) also improved
These are the largest weight-loss results yet reported for an injectable obesity drug in trials this size, exceeding what’s typically seen with tirzepatide or semaglutide
If the name retatrutide or “reta” sounds familiar to you, this injection is popular among the biohacking/peptide community. If Lilly is able to get FDA approval (which it plans to attempt in Q1 of 2027), then it will certainly become more mainstream. FDA approval requires a number of things, which include but are not limited to: proving the drug is safe, proving it works for the intended purpose, and showing it can be manufactured at scale
